Long-term nitrogen and phosphorus fertilization effects on N2 fixation rates and nifH gene community patterns in mangrove sediments

چکیده
The bioavailability of nutrients in sediments is one of the most important factors controlling ecological and physiological processes in the mangrove ecosystem (Alongi et al. 2002; Feller et al. 2002; Cheeseman & Lovelock 2004; Krauss et al. 2008). Although mangroves are highly productive, they are generally nutrient-deficient, relying on microbial and plant cycling processes for nutrients (Holguin et al. 2001; Feller et al. 2002; Reef et al. 2010). Anthropogenic activity in coastal areas can largely affect mangroves by external inputs of nutrients, causing negative changes in forest development and sediment geochemistry (Reef et al. 2010). The biological fixation of nitrogen is an important source for ‘new’ nitrogen in oligotrophic mangrove forests (Capone 1988; Lee & Joye 2006; Purvaja et al. 2008). Free-living diazotrophs are widely distributed within the

Phylogenetic Diversity of Diazotrophs along an Experimental Nutrient Gradient in Mangrove Sediments
The diversity of diazotrophs was studied in the sediment of mangrove forests (Twin Cays, Belize) subjected to a long-term fertilization with nitrogen and phosphorus. Terminal Restriction Fragment Length Polymorphism (TRFLP) and cloning of PCR-amplified nifH genes were combined via in silico analysis to assign clones to TRFLP-nifH phylotypes, as well as to characterize the occurrence of phylotyp...

We examined diazotroph activity in controlled experiments containing natural populations of nitrogen (N2)fixing microbes collected offshore from Kane’ohe Bay, Hawai’i, and from Station ALOHA (A Long-term Oligotrophic Habitat Assessment) in the North Pacific subtropical gyre. Quantitative polymerase chain reaction and reverse transcription-polymerase chain reaction approaches showed that the abu...

12 Benthic nitrogen (N2) fixation and sulfate reduction (SR) were investigated in the Peruvian 13 oxygen minimum zone (OMZ). Sediment samples, were retrieved by a multiple corer at six 14 stations (70 1025 m water depth) along a depth transect at 12°S, covering anoxic and 15 hypoxic bottom water conditions. Benthic N2 fixation was detected at all sites using the 16 acetylene reduction assay, wi...
